Experimental Validation

The in silico results were experimentally validated by real-time PCR. The study included a total of n=32 adult patients with severe obesity (BMI>35 kg/m2) who underwent bariatric surgery at the Hospital Sant Joan (Reus, Spain). SAT biopsies were obtained from obese males (n=8), obese females (n=8), obese males with T2D (n=8) and obese females with T2D (n=8). The study received ethical approval from the Ethics Committee of the Hospital Universitari Sant Joan (Reus, Spain) (EPIMET083/2018), and all participants signed the informed consent.

Figure 1: Gene expression-mediated validation of the in-silico results Gene expression-mediated validation of the in-silico results. Relative gene expression analysis in human SAT samples from obese and obese T2D males and females by qPCR. After obtaining informed consent, SAT samples were obtained during surgery, and RNA was extracted and used for real-time PCR. (A) Relative gene expression analysis for obesity in male and female obese samples. (B) Relative gene expression analysis for T2D in males (upper panel) and females (lower panel). In silico-analyses, fold-change results are shown for each gene. Each experimental group is represented by an n=8. Student’s t-test applied for significance: (*) p-value<0.05, (**) p-value<0.01.
